目的:探讨云芝糖肽在体外对细胞免疫和体液免疫功能调节的可能机制。方法分离健康人外周血单个核细胞(peripheral blood mononuclear cells,PBMCs),分为PBMCs空白组、云芝糖肽20μL与PBMCs共培养组和云芝糖肽40μL与PBMCs 共培养组,3组分别于培养24 h、48 h、72 h采用流式细胞仪检测淋巴细胞亚群的比例变化及T淋巴细胞表面CD3+/HLA-DR+、B淋巴细胞表面CD19+/CD69+表达情况,ELISA方法检测上清液中IgG水平。结果云芝糖肽与PBMCs共培养组CD4+T细胞比例明显上升(P<0.05),CD4+/CD8+细胞比值明显增高(P<0.05);云芝糖肽20μL与PBMCs共培养组T细胞表面CD3+/HLA-DR +,72 h时表达水平高于24 h(P<0.05);B细胞表面 CD19+/CD69+的表达显示从培养24 h到48 h表达升高而在72 h表达减少的趋势;PBMCs与云芝糖肽共培养2组上清IgG水平均明显高于PBMCs空白组(P均<0.05)。结论云芝糖肽在体外能促进CD4+T细胞比例上升及CD8+T细胞比例下降,提高CD4+/CD8+比值,并增加IgG分泌,具有一定的免疫调节作用。

목적:탐토운지당태재체외대세포면역화체액면역공능조절적가능궤제。방법분리건강인외주혈단개핵세포(peripheral blood mononuclear cells,PBMCs),분위PBMCs공백조、운지당태20μL여PBMCs공배양조화운지당태40μL여PBMCs 공배양조,3조분별우배양24 h、48 h、72 h채용류식세포의검측림파세포아군적비례변화급T림파세포표면CD3+/HLA-DR+、B림파세포표면CD19+/CD69+표체정황,ELISA방법검측상청액중IgG수평。결과운지당태여PBMCs공배양조CD4+T세포비례명현상승(P<0.05),CD4+/CD8+세포비치명현증고(P<0.05);운지당태20μL여PBMCs공배양조T세포표면CD3+/HLA-DR +,72 h시표체수평고우24 h(P<0.05);B세포표면 CD19+/CD69+적표체현시종배양24 h도48 h표체승고이재72 h표체감소적추세;PBMCs여운지당태공배양2조상청IgG수평균명현고우PBMCs공백조(P균<0.05)。결론운지당태재체외능촉진CD4+T세포비례상승급CD8+T세포비례하강,제고CD4+/CD8+비치,병증가IgG분비,구유일정적면역조절작용。
Objective It is to approach the mechanism of polysaccharopeptide ( PSP)on immune regulation in cellular and humoral in healthy humans peripheral blood lymphocytes in vitro .Mtehods Peripheral blood mononuclear cells ( PBMCs) from healthy donors were obtained by leukapheresis followed by ficoll -density centrifugation.The isolated cells were plated on 24-well plates (1000 μL per well) at a concentration of 1 ×106cells per milliliter in RPMI1640 supplemented with 10%fetal calf serum.The PBMCs were divided into three groups and were cultured for 72 hours in vitro.Flow cytometry was used to assess the percentages of human lymphocyte subsets.The expression of CD3 +/HLA-DR+T cells and CD19 +/CD69+B cells were measured by Flow cytometry at 24, 48 and 72 hours.The level of IgG in the supernatant was measured by enzymelinked immu-no-sorbent assay ( ELISA) at 24, 48 and 72 hours.Rseults After 72 hours of culture, the percentage of CD4 +T cells in PB-MCs+PSP groups were increased significantly as compared with those in PBMCs group (all P<0.05).Moreover, the ratio of CD4 +/CD8 +in PBMCs+PSP groups were significantly increased compared with those in PBMCs group (P<0.05).The ex-pression of CD3 +/HLA-DR+of T lymphocytes in PBMCs+PSP 20μL group at 72h was significantly higher at 24 hours (all P<0.05).PSP could obviously increase the level of IgG at 24, 48 and 72 hours.Conclusion The PSP can promote the rise of CD4 +and the decline of CD8 +, and increase CD4 +/CD8 +ratio and the secretion of IgG, so it have a certain role in im-mune regulation.
