目的:探测宫颈癌与白介素-8启动子区-251位点T/A单核苷酸多态性的相关性。方法:选取近2年确诊的78例宫颈癌患者和80例同年龄段健康女性血样,通过限制性片段长度多态性聚合酶链反应(PCR-RFLP)获得两组人群白介素-8启动子区-251位点的基因型频率和等位基因频率。结果:该位点经过 AseI 酶切结果获得3个基因型:AA、AT 和 TT。健康体检组人群中三种基因型频率分别为35%、46.25%和18.75%;宫颈癌组基因型频率分别为24.36%、46.15%和29.49%。宫颈癌组人群中T等位基因频率显著高于体检组(52.56% VS 41.87%,P<0.05)。结论:白介素-8启动子251位点的T/A多态性与宫颈癌相关,白介素-8基因可能是诱发宫颈癌肿瘤标志物。

목적:탐측궁경암여백개소-8계동자구-251위점T/A단핵감산다태성적상관성。방법:선취근2년학진적78례궁경암환자화80례동년령단건강녀성혈양,통과한제성편단장도다태성취합매련반응(PCR-RFLP)획득량조인군백개소-8계동자구-251위점적기인형빈솔화등위기인빈솔。결과:해위점경과 AseI 매절결과획득3개기인형:AA、AT 화 TT。건강체검조인군중삼충기인형빈솔분별위35%、46.25%화18.75%;궁경암조기인형빈솔분별위24.36%、46.15%화29.49%。궁경암조인군중T등위기인빈솔현저고우체검조(52.56% VS 41.87%,P<0.05)。결론:백개소-8계동자251위점적T/A다태성여궁경암상관,백개소-8기인가능시유발궁경암종류표지물。
Objective:To explore the relationship between -251A/T polynucleotide polymorphisms and cervical cancer at promoter region of interleukin 8.Methods:The blood sample of 78 cases of cervical carcinoma diagnosed in nearly 2 years and 80 cases of healthy women of the same age were selected,by restriction fragment length polymorphism polymerase chain reaction(PCR-RFLP),we obtained the genotype frequency and allele frequency of the -251 locus at promoter region of interleukin 8 of the two groups.Results:After AseI enzyme digestion,the site received 3 genotypes:AA,AT and TT.In the healthy group,three kinds of genotype frequencies respectively was 35% ,46.25% and 18.75% ;in cervical cancer group,genotype frequencies respectively was 24.36%,46.15% and 29.49%.In cervical cancer group,the frequency of the T allele was significantly higher than that in healthy group(52.56% VS 41.87% ,P<0.05).Conclusion:T/A polymorphism of the -251 locus at promoter region of interleukin 8 is associated with cervical cancer.Interleukin-8 gene may be a tumor marker inducing cervical cancer.
