目的:观察幽门螺杆菌重组热休克蛋白60对U937细胞分泌TNF-α与IL-17的影响。方法将由基因定点突变、重组后表达的胃癌来源株的幽门螺杆菌重组热休克蛋白60(rhsp60,即rhsp60组)及非胃癌来源株热休克蛋白60(hsp60,即hsp60组)分别与经佛波酯(PMA)诱导分化的U937细胞共培养,调整各组蛋白最终浓度分别为50、100、200μg/mL,并在12、18、24 h时收集上清液,ELISA检测上清液中TNF-α、IL-17水平,设磷酸盐缓冲液对照组。结果rhsp60组与hsp60组TNF-α、IL-17分泌水平均明显高于对照组,且同时间点、同浓度下rhsp60组明显高于hsp60组,差异均有统计学意义(P<0.05)。结论 rhsp60与hsp60均可促进U937细胞对细胞因子TNF-α、IL-17的分泌,且胃癌来源的rhsp60使TNF-α、IL-17分泌量更多。

목적:관찰유문라간균중조열휴극단백60대U937세포분비TNF-α여IL-17적영향。방법장유기인정점돌변、중조후표체적위암래원주적유문라간균중조열휴극단백60(rhsp60,즉rhsp60조)급비위암래원주열휴극단백60(hsp60,즉hsp60조)분별여경불파지(PMA)유도분화적U937세포공배양,조정각조단백최종농도분별위50、100、200μg/mL,병재12、18、24 h시수집상청액,ELISA검측상청액중TNF-α、IL-17수평,설린산염완충액대조조。결과rhsp60조여hsp60조TNF-α、IL-17분비수평균명현고우대조조,차동시간점、동농도하rhsp60조명현고우hsp60조,차이균유통계학의의(P<0.05)。결론 rhsp60여hsp60균가촉진U937세포대세포인자TNF-α、IL-17적분비,차위암래원적rhsp60사TNF-α、IL-17분비량경다。
Objective To investigate the effects of helicobacter pylori recombinant heat shock protein 60 on the cell se-cretion of TNF-αand IL-17 of U937 cells. Methods Hsp60 and rhsp60 were expressed by gene mutation and recombination , which were co-cultured with 50,100,200μg/mL of rhsp60(rhsp60 group) and hsp60(hsp60 group) with U937 cells induced by phorbol myristate acetate(PMA). The final concentration of the groups was adjusted as 50 μg/mL,100μg/mL,200μg/mL. The supernatant was collected at 12th,18th,24th h and IL-17,TNF-αsecretion was measured by ELISA. The experiment was setted the phosphate buffer as the control group. Results The TNF-α,IL-17 secretion concentrations in the hsp60 and rhsp60 groups were significantly higher than those in the control group. The TNF-α,IL-17 secretion concentrations in rhsp60 group was higher than that in hsp60 group at the same time and the same concentration. The difference had statistical significance (P<0.05). Conclusion The rhsp60 and hsp60 could promote the secretion of IL-17,TNF-αand rhsp60 derived from gastric cancer had more effects than hsp60 after stimulating U937 cells. secretory cell factor TNF-αand IL-17 of U937 cells.
