目的:观察畸胎瘤衍生生长因子( Cripto-1)在宫颈鳞癌组织中的表达变化,探讨其在肿瘤发生、发展过程中的作用。方法采用免疫组化法检测60例宫颈鳞癌组织、60例宫颈上皮内瘤变组织及24例正常宫颈组织中Cripto-1蛋白的表达,并分析宫颈癌组织中Cripto-1蛋白表达与临床病理参数及上皮-间质转化标记物( N-钙黏蛋白、波形蛋白、E-钙黏蛋白)的相关性。结果正常宫颈组织、宫颈上皮内瘤变组织、宫颈鳞癌组织中Cripto-1蛋白阳性表达率逐渐升高,且三者比较差异均有统计学意义(P均<0.05)。宫颈鳞癌不同临床分期及有无淋巴结转移患者Cripto-1蛋白表达阳性率比较均有统计学差异(P均<0.05);宫颈鳞癌患者Cripto-1蛋白表达阳性与N-钙黏蛋白、波形蛋白表达均呈正相关(r分别为0.561、0.328,P均<0.05),与E-钙黏蛋白表达呈负相关(r=-0.468,P<0.05)。结论 Cripto-1蛋白表达与宫颈鳞癌的发生、发展密切相关,通过上调Cripto-1蛋白表达调控上皮-间质转化以促进肿瘤的侵袭、转移。

목적:관찰기태류연생생장인자( Cripto-1)재궁경린암조직중적표체변화,탐토기재종류발생、발전과정중적작용。방법채용면역조화법검측60례궁경린암조직、60례궁경상피내류변조직급24례정상궁경조직중Cripto-1단백적표체,병분석궁경암조직중Cripto-1단백표체여림상병리삼수급상피-간질전화표기물( N-개점단백、파형단백、E-개점단백)적상관성。결과정상궁경조직、궁경상피내류변조직、궁경린암조직중Cripto-1단백양성표체솔축점승고,차삼자비교차이균유통계학의의(P균<0.05)。궁경린암불동림상분기급유무림파결전이환자Cripto-1단백표체양성솔비교균유통계학차이(P균<0.05);궁경린암환자Cripto-1단백표체양성여N-개점단백、파형단백표체균정정상관(r분별위0.561、0.328,P균<0.05),여E-개점단백표체정부상관(r=-0.468,P<0.05)。결론 Cripto-1단백표체여궁경린암적발생、발전밀절상관,통과상조Cripto-1단백표체조공상피-간질전화이촉진종류적침습、전이。
Objective To observe the expression changes of Cripto-1 in the cervical squamous cell carcinoma and to explore the effects on the tumor occurrence and development process.Methods Immunohistochemistry was used to exam-ine the expression levels of Cripto-1 in 60 cases of cervical squamous cell carcinoma, 60 cases of cervical intraepithelial ne-oplasia and 24 cases of normal cervical epithelia.The correlations of Cripto-1 expression with the clinical pathologic param-eters and the epithelial-mesenchymal transformation ( EMT) markers ( N-cadherin, Vimentin and E-cadherin) were also analyzed.Results The expression levels of Cripto-1 protein were gradually increased in the normal cervical epithelia, cervical intraepithelial neoplasia and cervical squamous cell carcinoma; and significant difference was found among these three groups (all P<0.05).The positive expression rate of Cripto-1 also showed significant differences in the cervical squamous cell carcinoma with or without lymph node metastasis and different clinical stages (all P<0.05).The positive expression of Cripto-1 was positively correlated with N-cadherin and Vimentin (r=0.561, r=0.328, respectively;all P<0.05), while it was negatively correlated with E-cadherin (r=-0.468, P<0.05).Conclusion The expression of Cripto-1 is closely related to the carcinogenesis and development of cervical squamous cell carcinoma, and may promote the invasion and metastasis of carcinoma by up-regulating the Cripto-1 expression to regulate the EMT.
