目的:探讨普伐他汀对兔动脉粥样硬化(AS)斑块稳定性及血清脂蛋白相关磷脂酶A2(Lp-PLA2)表达的影响。方法取大白兔24只,随机分为甲、乙、丙三组,每组8只。甲组不造模,乙、丙组采用球囊拉伤法制造AS模型,丙组给予普伐他汀干预,然后观察其病理改变及血清Lp-PLA2表达情况。结果与甲组比较,第5、10周乙、丙组大白兔血清Lp-PLA2水平显著增高,差异均有统计学意义(P<0.05),且第10周乙组大白兔血清Lp-PLA2水平增高更为明显。与乙组比较,第5、10周丙组大白兔血清Lp-PLA2水平均明显下降,差异均有统计学意义(P<0.05)。第5、10周乙组大白兔血清Lp-PLA2水平均显著高于实验前,差异均有统计学意义(P<0.01)。第5、10周丙组大白兔血清Lp-PLA2水平略高于实验前。普伐他汀干预后AS斑块的病理学形态、结构均发生改变,斑块结构更加趋于稳定。结论普伐他汀能降低AS动物的Lp-PLA2水平,促进斑块稳定。

Objective To investigate the influence of pravastatin on the atherosclerotic plaque (AS) stability and serum Lp-PLA2 expression in rabbit. Methods 24 rabbits were randomly divided into the group A,B and C,8 cases in each group. The group A was non-model. The group B and C adopted the balloon pulling injury method for establishing the AS model. Then the pathological changes and serum Lp-PLA2 expression were observed. Results Compared with the group A,serum Lp-PLA2 levels at 5,10 weeks in the group B and C were increased significantly,the differences were statistically significant(P<0.05),moreover the increase of serum Lp-PLA2 level at 10 weeks in the group B was more obvious. Compared with the group B ,the serum Lp-PLA2 levels at 5,10 weeks in the group C was significantly decreased,the difference was statistically significant(P<0.05). The serum Lp-PLA2 levels at 5,10 weeks in the group B were significantly higher than those before experiment,the difference was sta-tistically significant(P<0.01). The serum Lp-PLA2 levels at 5,10 weeks in the group C were slightly higher than those before ex-periment. The pathological morphology and structure of AS plaque after pravastatin intervention were changed and the plaque structure trended to be more stable. Conclusion Pravastatin can reduce Lp-PLA2 level in the AS animal and promotes the AS plaque stability.
