目的:将传统中医辨证方法与现代系统生物学理论相结合,研究2型糖尿病中医辨证分型及虚实证与基于超高效液相色谱法(UPLC)-四极杆飞行时间(QTOF)-质谱(MS)/MS的血清代谢组学的关系。方法选择2013年6月至2014年4月住院并确诊为2型糖尿病患者90例(辨证分为阴虚热盛、气阴两虚、血淤气滞3种证型)。应用UPLC-QTOF-MS/MS方法测定健康者及不同中医证型2型糖尿病患者血清内源性代谢物,并用偏最小二乘-判别式分析(PLS-DA)代谢谱。结果 PLS-DA分析结果表明,该法不仅可有效区分疾病组和健康对照组,也可明显区分患者中阴虚热盛、气阴两虚、血淤气滞3种证型。结论基于UPLC-QTOF-MS/MS和模式识别的代谢组学技术在揭示传统中医理论本质及对2型糖尿病的临床研究方面具有广泛的应用前景。

Objective To study the relationship between the traditional Chinese medicine syndrome differentiation and excess or deficiency syndrome in type 2 diabetes mellitus (T2DM) with the serum metabolomics based on UPLC-QTOF-MS/MS by combining the traditional Chinese medicine differentiation method with the modern systematic biological theory. Methods 90 inpatients diagnosed as T2DM in our hospital from June 2013 to April 2014 were selected and divided into 3 kinds of syndrome type including the type of Yin deficiency and intense heat ,type of deficiency of both Qi and Yin and type of Qi stagnancy and blood stasis. The endogenous metabolites in the healthy individuals and different Chinese medicine syndrome types of T2DM were detected by the UPLC-QTOF-MS/MS method. Then the metabolic profiling was analyzed by using the partial least squares dis-criminant(PLS-DA) method. Results The PLS-DA analysis results showed that this method not only effectively distinguished the disease group from the healthy control group,but also differentiated the three syndrome types of Yin deficiency and intense heat, deficiency of both Qi and Yin,and Qi stagnancy and blood stasis in T2DM patients. Conclusion The metabolomic technique based on UPLC-QTOF-MS/MS and pattern recognition has the extensive application prospects in revealing the essence of tradi-tional Chinese medicine and clinical research of T2DM.
